5-HTP is used to make both serotonin and melatonin. Serotonin is a neurotransmitter that also acts as a hormone. In the brain, serotonin influences mood, sleep, cognition, and appetite. As the primary neurotransmitter in the gastrointestinal (GI) tract, serotonin governs digestion speed and GI motility.

Serotonin plays essential roles in regulating mood. It allows you to feel joy and calm. As a precursor to melatonin, it is central to your body’s sleep-wake cycle. It also supports the appetite center of your brain. Here, it is responsible for feelings of satiety and satisfaction. Healthy serotonin levels may also support normal regulation of appetite and cravings.

Serotonin synthesis is dependent on the availability of 5-HTP. However, 5-HTP is not abundant in a typical diet. Instead, your body manufactures 5-HTP from the amino acid L-tryptophan.

Both L-tryptophan and 5-HTP are considered serotonin precursors. When working to increase serotonin levels, supplementation with 5-HTP is preferable. This is because tryptophan is readily diverted to other functions in the body, such as niacin and protein production.

Tryptophan deficiency may be quite common. It is the least abundant amino acid in the human diet. Its absorption can be easily impaired from poor digestion. In addition, tryptophan-containing foods do not significantly increase 5-HTP levels.

Why 5- HTP Works

Serotonin (5-hydroxytryptamine or 5-HT) is your neurotransmitter responsible for feelings of joy and calm. The direct chemical precursor to serotonin is L-5-hydroxytryptophan (5-HTP). This is made from the amino acid L-tryptophan.

The production of serotonin depends on the availability of L-tryptophan. Unfortunately, it is challenging to get enough tryptophan into your body. Tryptophan is the least common amino acid in the human diet. Many people are lucky to bring in between 0.5 and 1.5 grams per day. Low protein diets, insufficient stomach acid, acid-blocking medications, and digestive disorders can all lead to tryptophan deficiency.

Once tryptophan from foods is digested and absorbed, your body must transform it into 5-HTP. This requires the cofactor tetrahydrobiopterin (BH4). A deficiency in BH4 can lower conversion rates and 5-HTP levels.

In addition to dietary challenges, tryptophan is readily ‘stolen’ away to be used in other biochemical processes. Niacin deficiency, cellular stress, and low antioxidant levels can all play a role in stealing tryptophan and lowering 5-HTP levels.

Over 95 percent of your body’s L-tryptophan is used in the kynurenine pathway. This biochemical pathway produces a niacin-derived compound called nicotinamide adenine dinucleotide (NAD+). This critical coenzyme is found in every cell where it is involved in hundreds of metabolic processes.

When niacin levels are low, your body activates the IDO and TDO enzymes. This draws tryptophan down the kynurenine pathway to make niacin and ultimately NAD+. It takes approximately 60 tryptophans to make one niacin. As you can see, producing niacin via the kynurenine pathway can play a huge role in lowering tryptophan levels. Inflammation, stress, and infections can all activate IDO and TDO enzymes and steal tryptophan down this pathway.

When a cell is low in antioxidants, the enzyme tryptophan hydroxylase (TPH) slows down. This enzyme converts tryptophan into 5-HTP. The TPH enzyme relies on BH4 as a cofactor. This cofactor is incredibly sensitive to oxidative stress. With low levels of antioxidant nutrients (such as vitamin C), BH4 levels can fall and slow the production of 5-HTP.

By supplementing with 5-HTP, you bypass the enzymatic steps involved in converting tryptophan to 5-HTP. 5-HTP is not stolen away for other biological processes as tryptophan is. 5-HTP supplementation has been shown to support healthy serotonin levels. In turn, this supports emotional well-being, a healthy appetite, and cognitive health. As the precursor to melatonin, 5-HTP also supports healthy sleep and circadian rhythms.

Sleeping like a baby

This really helps me fall asleep and stay asleep. I kept struggling with the midnight stare at the ceiling. This has helps put that to an end! Thankfully!
